Does an Increase in Temperature Influence the Short-term Toxicity of the Pharmaceutical Fluorouracil to Daphnia magna?

In the present scenario of global warming and pollution increase, it is very important to investigate the effects of temperature increase on the toxic effects of environmental contaminants, such as pharmaceuticals that are considered emergent contaminants of high concern. Among pharmaceuticals, those used as chemotherapeutic agents are of special concern due to their biological activity. Fluorouracil (5-fluorouracil, hereafter indicated as 5-FU) is a chemotherapeutic medicine widely used in the treatment of various neoplasms that is also used in the treatment of several dermatologic problems. As result of its wide use, 5-FU may reach aquatic ecosystems where it can cause adverse effects on aquatic life. The goal of the present study was to investigate the effects of temperature rise (from 20ºC to 25ºC) on the short-term toxicity of 5-FLU to Daphnia magna, a freshwater crustacean that is widely used in Ecotoxicology. Two D. magna bioassays were carried out in laboratory conditions: one at 20ºC and the other at 25ºC. In each bioassay, D. magna juveniles were exposed to several concentrations (ppb - ppm range) for 96h hours with no food. The effect criteria was juvenile mortality recognised through the immobilization. The results indicate that the temperature rise from 20ºC to 25ºC increased the short-term toxicity of 5-FLU to D. magna. The type of interaction between temperature rise and 5-FLU in D. magna juveniles was determined, and its implications will be discussed.
